The basic principle of time dependent single photon counting is to excite the sample with a narrow light pulse, and then detect the time when the first fluorescent photon emitted by the sample reaches the optical signal receiver. TAC proportionally converts this time into corresponding voltage pulses, and then converts this electrical pulse into a multi-channel analyzer through AD conversion. In the multi-channel analyzer, these output pulses are sequentially sent to each channel for accumulation and storage. A histogram that is consistent with the original waveform is obtained. The probability of detecting photons within a certain time interval is directly proportional to the fluorescence emission intensity, and the decay pattern of fluorescence intensity is obtained by repeated measurements.
